optimal inoculum levels for the resistance screening of sugar beet to root- knot nematode under greenhouse condition

چکیده
sugar beet (beta vulgaris) is one of the hosts for different root-knot nematode species. in this study, firstly the effect of different inoculum levels including 250+250, 500, 500+250, 750, 500+500 and 1000 second stage juveniles of meloidogyne javanica were evaluated on a susceptible variety named jolgeh. the results showed that all inoculum levels were suitable for infection on sugar beet plants and that the gall numbers at one and two time inoculations were not statistically different (p≥0.01). therefore, 500 second stage juvenile of root-knot nematode was the lowest inoculum level that could be used to infect sugar beet plants in the greenhouse. in the second experiment, the best treatment was applied on jolgeh (susceptible to root-knot nematode) and pauletta (commercial resistant variety to cyst nematode) varieties and also seven half-sib families derived from sb33 population (carrying root-knot nematode resistance gene), to evaluate its generalization for screening of resistant/susceptible genotypes. the total number of root galls was taken as nematode resistance criterion. results showed that jolgeh and pauletta varieties with average number of more than 100 galls were considered as highly susceptible varieties whereas the seven half-sib families with average number of 10 galls (

development of an snp marker for sugar beet resistance/susceptible genotyping to root-knot nematode
linked and/or gene-based molecular markers have been used widely in marker-assisted selection (mas) to differentiate resistant and susceptible genotypes. resistance to meloidogyne spp. in beta vulgaris l. is mediated by a single dominant gene (r6m-1). using allele-specific primers (asps), an snp marker harboring a single nucleotide polymorphism (a/g), linked to the resistance gene was developed...
متن کاملevaluation of resistance of some sugar beet genotypes to beet cyst nematode under greenhouse conditions
resistance of 11 genotypes of sugar beet to cyst nematode was evaluated in two separate greenhouse experiments in completely randomized design. in each experiment, 50 seedlings of each genotype were inoculated with 1000 nematode larves in several times. after 9 weeks, the number of cysts on each plant was counted. data were analyzed with sas program. the results showed that the genotypes differ...
متن کاملevaluation of superior sugar beet genotypes for resistance to important root rot pathogens in the greenhouse
four sugar beet genotypes with different reactions to root rot pathogens were evaluated for resistance to important root rot agents including pythium aphanidermatum, phytophthora drechsleri, rhizoctonia solani ag-2-2 and r. solani ag-4. inoculums for pythium and phytophthora were 3-5 day actively growing colony of the pathogens on cma medium, while for rhizoctonia isolates were nine infected co...
